A lectin with molecular mass around 200 kDa was isolated from the serum of the Indian catfish Clarias batrachus. The bioactivity of this serum lectin was Ca^2^+ and pH dependent. The lectin appeared to be specific for @a-methyl galactose and sialoglycoproteins like porcine and bovine submaxillary mucin and could agglutinate human, rabbit, mice, rat and chicken erythrocytes. This fish lectin was able to specifically agglutinate different gram negative bacteria. When it was checked against different strains of the fish pathogen Aeromonas sp., it significantly altered the viability and pathogenicity of the bacteria. Binding of the lectin to Aeromonas sp., resulted in a dose dependent increase in the bactericidal activity of fish macrophages. However, when the lectin was checked against different gram positive bacteria it could not agglutinate or affect the viability of those strains and also failed to bring about any significant change in the bactericidal potential of fish macrophages. The lectin was able to induce the proliferation of head kidney lymphocytes of Clarias and helped in the release of 'IL-1' like cytokines from head kidney macrophages.

The concentrations of total mercury (THg), stable isotopes of nitrogen (@d^1^5N) and carbon (@d^1^3C), and the diet of the African sharptooth catfish Clarias gariepinus in Lake Awassa, Ethiopia, were studied from January 2003 to February 2004. Values of the @d^1^5N were used as an index of trophic position in four length groups and compared to actual stomach contents. The diet of C. gariepinus within the length range of 201-600 mm L"T mainly consisted of the small barb Barbus paludinosus, aquatic insects, and mollusks. The proportion of fish prey in the diet was 60% by volume, irrespective of fish size. The differences in @d^1^5N values of individuals within and between length classes were less than 3%%, and were not significantly related to total length, showing the similarity in trophic niche of the different sizes, which corresponded to recorded stomach contents. Mercury concentrations were in the range of 0.002-0.154 mg kg^-^1 ww, and had no significant relationship to total length. Hence, even large specimens of C. gariepinus have Hg values below the WHO threshold of 0.2 mg kg^-^1 ww. The slope of the regression line between log [Hg] and @d^1^5N was small, 0.06, indicating the absence of trophic shifts and biomagnification of Hg in larger specimens in our samples. The low Hg concentrations in C. gariepinus compared to the Hg concentrations in other piscivorous fish species in Lake Awassa, such as Barbus intermedius and B. paludinosus, may be due to its dependence on invertebrate preys at small size, diet switching towards low Hg prey fish at larger size, and growth biodilution owing to higher growth rate.

We studied the hepatocellular alterations induced by sub-lethal concentrations (0.50 @mM) of arsenic in Indian catfish Clarias batrachus L. Sub-lethal arsenic exposure altered serum aspartate aminotransferase and alkaline phosphatase levels and brought about significant changes in different serum biochemical parameters. Arsenic exposure reduced total hepatocyte protein content and suppressed the proliferation of hepatocytes in a time-dependent manner. Routine histological studies on liver documented arsenic-induced changes characterized by dilated sinusoids, formation of intracellular edema, megalocytosis, vacuolation and appearance of hepatic cells with distorted nuclei. Transmission electron microscopy of hepatocytes further revealed hyperplasia and hypertrophy of mitochondria, development of dilated rough endoplasmic reticulum and changes in peroxisome size with duration of arsenic exposure. Degeneration of mitochondrial cristae and condensation of chromatin was also evident in arsenic-exposed hepatocytes. A significant number of hepatocytes isolated from arsenic-exposed fish stained with annexin V and demonstrated DNA ladder characteristic of apoptosis. Single-cell gel electrophoresis of exposed hepatocytes also revealed the development of comets usually seen in apoptotic cells. Using specific inhibitors it was determined that the arsenic-induced apoptosis of hepatocytes was caspase-mediated, involving the caspase 3 pathway.

We studied the effects of a sublethal concentration of pyrethroid insecticide fenvalerate on metabolic enzymes, RNA and protein of brain, liver and skeletal muscle of the freshwater catfish, Clarias batrachus. Exposure to fenvalerate gradually decreased the activity of citrate synthase (CS), glucose 6-phosphate dehydrogenase (G6-PDH) and lactate dehydrogenase (LDH) in brain, liver and skeletal muscle up to 21 days. The maximum decrease in enzyme activity was 23-47%. Withdrawal of fenvalerate from the medium for 21 days restored enzyme activity to their control level in all three tissues. RNA and protein content in brain, liver and skeletal muscle decreased significantly with exposure of fenvalerate up to 21 days. The maximum decrease in RNA and protein was 22-32%. Withdrawal of fenvalerate from the medium for 21 days restored the RNA and protein contents to control levels. The present study suggests that fenvalerate impairs cellular metabolism and its biochemical effects are reversible after withdrawal of fenvalerate.
